Abstract
To establish an efficient in vitro regeneration protocol for orchardgrass (Dactylis glomerata L.), type of medium, several plant growth regulators and their various concentrations were evaluated in a factorial experiment for callus induction and plant regeneration. Endosperm supported mature embryo were used as explants and optimum response in callus induction was observed on Chu medium with 12 mg/L dicamba (3.6-dichloro-2-methoxybenzoic acid). The highest responding embryogenic callus (15%) was found with Murashige and Skoog medium and 8 mg/L 2.4-D (2,4-dichlorophenoxy acetic acid). The highest regeneration efficiency was achieved on MS medium (17 plantlets) with 8 mg/L 2,4-D. Endosperm-supported mature embryos of orchardgrass can be successfully used as explants. Therefore, this protocol provides a basis for future studies on genetic improvement and in vitro mutation studies.
