Increasing the Efficiency of in vitro Chromosome Doubling and Ploidy Determination in Haploid Sugar Beet (Beta vulgaris L.)

Abstract

This research was carride out in Denmark in 1990-1991. To investigate the effect of different colchicine substrates to chromosomal doubling and to determinate of ploide Ievel of colchicine treated-in vitro seedling. The haploid sinlge shoots derived from 4 sugar beet (Beta vulgaris L.) Iines (H890018. H890021, H890535 and H891538) were cultivated on 8 differentcolchicine substrates after being kept in darkness for 3-4 days. The single shoots were transferredwing 3, 4 and 5 days colchicine substrate treatments. The increasing durations of the colchicine substrate treatment raised both doubling and mortality rates of shoots. respectively 8.5. 13.3 and 23.0. and 27. 7 . 32.4 and 39.5 % for 3, 4 and 5 days procedure. Doubling and mortality on the colchicine unterated shoots were 0 and 8.8 % respectively. Average 14.5 % of doubling and 14.2 % of unrooted were observed from all shoots. Ploidy Ievels of plantlets were determined in vitro and in vivo by flow cytometry. In vitro determination of the true ploidy Ievels of the shoots mostly raised up to 77. 7 % average compared to those of in vivo.
