Polymorphism and Phylogenetic Relations Among Turkish Species in the Genus Sternbergia (Amaryllidaceae) As Determined by RAPD-PCR

Abstract
We used Random Amplified Polymorphic DNA Polymerase Chain Reaction (RAPD-PCR) to identify Sternbergia Waldst. & Kit. species. Two primers of random sequence are used at a low annealing temperature. These primers were useful in identifying six species of Sternbergia. Reaction with two primers, L1 and L2, produced amplified DNA bands whose size allowed the seperations of four species: S. lutea (L.) Ker-Gawl. ex Sprengel and S. sicula Tineo ex Guss. from S. colchiciflora Wadst. & Kit. S. fischeriana (Herbert) Rupr., S. candida Mathew & Baytop, and S. clusiana. (Ker-Gawl) Ker-Gawl. ex Sprengel. In contrast. S. lutea and S. sicula were found to be almost indistinguishable.
