Determination of some Biochemical Properties of Wheat (Triticum aestivum L.) \alpha-Amylase Enzyme and Effects of Endosulfan on Enzyme Activity
Abstract
In this study, some of the biochemical properties of \alpha-amylase enzyme, which plays an important role in seed germination were determined in wheat grain. The effects of endosulfan on enzyme activity of \alpha-amylase and the total protein amount of wheat grain were also investigated. Wheat (Triticum aestivum Pehlivan) \alpha-amylase enzyme activity was obtained at highest Ievel between 2nd and 5th days of germination. The optimum conditions for this enzyme were determinated at pH: 5.2, incubation temparature. 30°C, incubation period: 3 minutes. It was also determinated that the Iiquid and powder forms of endusulfan, depending on dose, inhibited the enzyme activity and caused decreases in the total amount of protein. The reason could be the inhibition effect of endosulfan on the enzyme synthesis of \alpha-amylase.
