Abstract
Background/aim: Persistent inflammation disrupts keratinocyte migration and contributes to delayed wound healing. Lipopolysaccharide (LPS)-induced inflammatory models are widely used to investigate impaired reepithelialization and inflammatory cellular dysfunction. Resveratrol, a naturally occurring polyphenolic compound, exhibits antiinflammatory and antioxidant properties; however, its therapeutic effects on established inflammatory dysfunction remain insufficiently characterized. This study investigates the effects of resveratrol on LPS-induced keratinocyte dysfunction and compares them with those of boric acid (BA) under inflammatory conditions.
Methods: Human HaCaT keratinocytes were exposed to 10 μg/mL LPS to establish an inflammatory microenvironment. Following LPS pretreatment, the cells were treated with resveratrol (2, 5, or 10 μM) or BA (20 or 100 μM). Cellular metabolic activity was evaluated using the Cell Counting Kit-8 (CCK-8) assay, and cell migration using a scratch wound-healing assay. Oxidative stress-related biochemical parameters were also evaluated. For the statistical analysis, the Kruskal–Wallis test was used, followed by Dunn’s post hoc test for multiple comparisons.
Results: LPS exposure significantly impaired keratinocyte migration, reducing wound closure rates from approximately 60% in control cells to nearly 35% in LPS-treated cells. Resveratrol treatment at 5 and 10 μM was associated with higher mean wound closure rates than LPS treatment alone, although the differences were not statistically significant. Similarly, low-dose resveratrol and boric acid also failed to improve keratinocyte migration under inflammatory conditions to any significant degree. While resveratrol alone reduced the CCK-8 metabolic signal, cells treated with resveratrol following LPS conditioning maintained metabolic activity levels comparable to the controls, suggesting an absence of overt cytotoxicity. Resveratrol also modulated oxidative stress-related responses, as evidenced by reductions in malondialdehyde levels and enhanced antioxidant enzyme activity.
Conclusion: In the present study, resveratrol was associated with improved keratinocyte migration under LPS-induced inflammatory conditions. These findings suggest that resveratrol may be a promising modulator of inflammatory wound microenvironments, which are characterized by impaired re-epithelialization, oxidative stress, and persistent inflammatory signaling.
